Plicamycin may also be used for purposes other than those listed in these treatment guidelines.

The doctor will determine the appropriate amount and frequency of treatment with plicamycin depending on the type of cancer being treated and other factors. What is the dose of Plicamycin for adults?

Adult dosage for testicular cancer

For the treatment of patients with carefully selected malignant tumors of the hospitalized testicles that are unlikely to be cured by surgery and / or radiation: 25-30 mcg (0.025-0.030 mg) per kilogram of body weight. Therapy should be continued for a period of 8-10 days if there are no significant side effects or toxicity during therapy. A course of therapy consisting of more than 10 daily doses is not recommended. Individual daily doses should not exceed 30 mcg (0.030 mg) per kilogram of body weight.

In patients with responsive tumor, some degree of tumor reduction is usually seen within 3-4 weeks after the initial therapy sequence, additional therapy sequences at monthly intervals are guaranteed. When the tumor is drastically reduced, it is recommended that additional courses of therapy be given at monthly intervals until the tumor disappears completely or until the tumor develops or new tumors appear even though the course of therapy is continued.

Usual adult dosage for hypercalcemia

In hypercalcemia associated with the development of a malignant tumor, the recommended dose of the drug is 25 mcg (0.025 mg) per kilogram of body weight per day for 3-4 days.

If the hypercalcemia has not subsided during the initial course of therapy, additional courses of therapy may be given at intervals of 1 week or more to achieve the desired results or to maintain normal serum calcium and urinary calcium excretion. Normal calcium balance may be maintained with a single dose per week or on a schedule of 2-3 doses per week.

Safety and effectiveness have not been confirmed in pediatric patients (less than 18 years).

In what dosages and preparations is Plicamycin available?

Mithracin (plicamycin) is available in a vial as a freeze-dry preparation for intravenous administration. Each bottle contains 2500 mcg (2.5 mg) of Mithracin (plicamycin) with 100 mg of mannitol and enough phosphate disodium to adjust to a pH of 7.

To dilute, add 4.9 mL of sterile water for injection aseptically to the contents of the bottle and shake the bottle to dissolve it. Each mL of the solution will then contain 500 mcg (0.5 mg) of Mithracin (plicamycin).

What to do in an emergency or overdose?

In case of an emergency or overdose, contact the local emergency services provider (118/119) or immediately to the nearest hospital emergency department.

What should I do if I forget to take medicine or forget to take medicine?

If you forget a dose of this medicine, take it as soon as possible. However, when it is nearing the time of the next dose, skip the missed dose and return to the usual dosing schedule. Don't double the dose.
